ICSI involves injecting a single sperm cell into a matured oocyte, culturing the injected egg until it divides, with the intent to produce a viable embryo. This innovative technique has a high chance of fertilization, despite the sperm quality or quantity. ICSI is often the preferred route for rare and high-performance horses where horse owners and breeders hope to help the genetics live on through many generations to come.

THE ICSI PROCESS

Turning Possibility Into Life

STEP 1

Collection

Oocytes are extracted from the mare.

STEP 2

Maturation

The oocytes mature in a safe and secure environment.

STEP 3

Sperm Sample

A sperm sample is taken.

STEP 4

Injection

The sperm cell is inserted into the oocyte.

STEP 5

Embryo Culture

The oocyte will spend the next few days culturing to become an embryo.

STEP 6

Transfer

The embryo can be transferred to a mare or stored for the future.

THE ICSI PROCESS

Thawing Protocol for Frozen Embryos-Cryolock Straw

Warm all media to 38C (Media 1 and Media 2 – provided by Viagen Equine, standard embryo transfer media – supplied by veterinarian)

Make a thaw plate using a 4 well plate with roughly 1 ml of Media 1 in well 1, roughly 1 ml of Media 2 in well 2. Leave thaw plate on warming plate set at 37C.

Remove cap from cryolock while holding under LN2 using hemostats or other tool to hold just above the end of the cap. (If you grab too high you can crush cryolock cap and tip with embryo)

Pull cryolock out of LN2 and quickly plunge tip into Media 1 in well 1.

Pause for 3 seconds, then gently shake cryolock, verify visually that embryo is in well 1.

Hold embryo in Media 1 for 1 min.

Move embryo into Media 2 in well 2 and incubate for 5 min.

Towards the end of the 5 min incubation, add 1 ml of your standard embryo transfer media (supplied by veterinarian) to well 3.

At the end of 5 min incubation in well 2, move embryo into well 3.

Embryo is ready for immediate loading and transfer. If more than 5 minutes is needed prior to loading and transfer, temperature on warming plate should be reduced to 34C and well 3 should be covered with Ovoil.
